What is SYK Signalling?
Key Points. Spleen tyrosine kinase (SYK) is a non-receptor tyrosine kinase that has long been thought to exclusively mediate signalling by receptors of the adaptive immune response (B cell receptors (BCRs) and Fc receptors (FcRs)).
What does SYK do?
Spleen tyrosine kinase (SYK) has been known to relay adaptive immune receptor signalling. However, recent reports indicate that SYK also mediates other, unexpectedly diverse biological functions including cellular adhesion, innate immune recognition, osteoclast maturation, platelet activation and vascular development.
How is SYK activated?
SYK is recruited by interaction with ppITAMs, and undergoes autophosphorylation (34, 56) as well as phosphorylation by membrane-associated SRC kinases (7, 8). These phosphorylation events activate SYK, initiating multiple signaling cascades, which results in an array of inflammatory or immunological outcomes (9).
What is ZAP70?
The ZAP70 gene provides instructions for making a protein called zeta-chain-associated protein kinase. This protein is part of a signaling pathway that directs the development of and turns on (activates) immune system cells called T cells. T cells identify foreign substances and defend the body against infection.
What are Syk inhibitors?
Syk inhibitors are a new group of small molecule inhibitors targeting BCR-mediated signaling pathways. The second generation compound, entospletinib, showed promising results in clinical trials against B-cell malignancies, mainly CLL.
What is Syk inhibitor?
What are Btk inhibitors?
An orally bioavailable, selective inhibitor of Bruton’s tyrosine kinase (BTK), with potential antineoplastic activity. Upon administration, M7583 targets and covalently binds to BTK, thereby preventing its activity.
Does casein have a SH2 domain?
The SH2 (Src Homology 2) domain is a structurally conserved protein domain contained within the Src oncoprotein and in many other intracellular signal-transducing proteins….SH2 domain.
